PREPARATION OF BLOOD-PLASMA KALLIKREIN AND DETERMINATION OF BLOOD-PLASMA PREKALLIKREIN

PURPOSE:To prepare the titled compound useful as a medicine for cardio-vascular system, easily, by adding an activating agent such as kaolin to the blood-plasma of mammals adjusted to a specific pH value, thereby activating the prekallikrein and at the same time adsorbing the activated kallikrein to the activating agent, and then eluting the adsorbed kallikrein with a specific solution. CONSTITUTION:Human or mammalian blood-plasma is mixed with a buffer solution [e.g. equal volume of 0.1M phosphate buffer solution (pH 6.0), etc.] to adjust the pH of the mixture of to 6.5. An activating agent such as kaolin or Celite is added to the mixture and stirred thoroughly at a low temperature to activate the prekallikrein. The amount of the activating agent is =15mg/1ml of the blood- plasma. The activated kallikrein is simultaneously adsorbed to the activating agent. The adsorbed kallikrein is eluted with a protamine solution prepared by dissolving 0.5-4mg of protamine in 1ml of 0.02M tris-hydrochloric acid buffer solution (pH 8.0) to obtain the blood-plasma kallikrein in high efficiency. The kallikrein plays an important role in quinine, coagulation or adenolytic system for maintaining the homeostatis.
