A therapeutic compsn. is prepd. by incubating a medium contg. messenger RNA (I); an accelular extract contg. components necessary for protein synthesis obtd. from human, animal, vegetable or bacterial cells and a buffer to maintain pH 6-9. The medium also contains (pref. ranges) stabilising monovalent (50-100 mM) and divalent (1-5 mM) ions; a reducing agent (ii; 1-5 mM); a polyamine (III; 0.1-0.3 mM); a mixture of the 20 natural amino acids (20-50 mu M); ATP (0.5-2 mM); GTP (10-50 mu M) and products (V) which generate nucleoside triphosphates (5-15 mM). incubation is at 10-40 degrees C, esp. about ambient. USe/ ADVANTAGE : ADVANTAGES The compsns. have interferon activity (formed by 'translation' of (I) and hence specific for the species from which (I) is derived), and any type of interferon can be produced in high yield. They are useful as antivirals, immunodepressants and to prevent development of metastases in cancerous states esp. in sarcoma of the bone.
