3-Hydroxyrifamycin S (I), 3, 31-dihydroxyrifamycin S (II), and l-deoxy-l-oxarifamycin S (III) were produced by fermn. with Nocardia mediterranei strain obtained from streptomyces mediterranei ATCC l3685. Thus, a preculture of S. mediterranei was inoculated into 3L of pH 7 medium contg. glucose 22, beef ext. 5, peptone 5, yeast ext. 5, casein hydrolysate 3, and NaC1 l.5g/L and incubated in shaken flasks at 28≦̸C for 7 days. The broth was then filtered and extd. with CHCL3. Finally, the ext. was concd. and chromatographed on silica gel, eluting stepwise with increasing concns. of MeOH in H2O. I, II, and III were thus sepd. and further purified by TLC.
