DNA FRAGMENT CONTAINING STRUCTURAL GENE OF NEUTRAL PROTEASE AND PRODUCTION OF NEUTRAL PROTEASE

PURPOSE:To obtain a DNA fragment giving a neutral protease useful for the preparation of foods, etc., by using a bacterium belonging to Bacillus genus as a chromosome donor, incising the chromosome, joining the obtained DNA fragment to a vector, and transforming a host bacterium using the resultant recombinant DNA. CONSTITUTION:A bacterium belonging to Bacillus genus, e.g. Bacillus amyloliquefaciens, etc. is used as a chromosome donor, and its chromosome is incised preferably with the restriction enzyme, Sau 3Al, to obtain a DNA fragment. The fragment is joined to a vector, and the obtained recombinant DNA is used for the transformation of a host bacterium cell. A chromosome fraction which is a DNA fragment containing the structural gene of a neutural protease gene as well as a promotor range and signal sequence necessary for its manifestation and secretion can be selected by the above genetic engineering process.
