REAGENT FOR MEASURING ALPHA2 -MACROGLOBULIN BOUNDABLE CALLICREIN ACTIVITY

PURPOSE:To enable measurement of the alpha2-macroglobulin bondable callicrein activity in a human bodily fluid by using an insolubilized carrier formed by covalent bond of an anti alpha2-macroglobulin antibody and a substrate specific to callicrein as a reagent. CONSTITUTION:An insolubilized antibody formed by sensitizing the alpha2-macroglobulin (alpha2-MG) refined from a human blood plasma to a house rabbit, and obtaining the refined anti alpha2-MG antibody refined from the antiserum thereof by covalent bond or physical adsorption together with a polystyrene bead, etc. is prepd. Said antibody and a synthetic substrate L-prolyl-L-phenylaranyl-L-alginyl- 4-methylcumaryl-7-amide having high specificness with callicrein are brought into reaction with the blood plasma to be examined for about one hour at about 37 deg.C, and the fluorescent intensity is measured at 380nm excitation wavelength and 440nm fluorescence wavelength. The activity of the alpha2-MG.callicrein conjugate in the blood plasma is thus measured with good accuracy and is made useful in the diagnosis of the heptocirrhosis and subacute hepatitis.
