PURPOSE:To determine quantitatively human chorionic gonandotropin (HCG) with high sensitivity by using a solid phase prepd. by bonding a specific antibody or the fragment of the antibody to HCG and the specific antibody fragment for HCG bonded with enzyme. CONSTITUTION:An IgG fragment is sampled by ammonia sulfate fractionation and DEAE cellulose chromatography from the antiserum obtd. by immunizing a house rabbit and is brought into reaction with hepsin to obtain an Fab' fragment. The solid phase prepd. by bonding the fragment with a polystyrene ball (4mm. diameter), etc. is prepd. On the other hand, a composite body formed by bonding the peroxidase originating in a wester horse raddish with the same Fab' fragment is prepd. The solid phase and the composite body are brought into reaction with an HCG-contg. sample and the enzyme activity of the composite body bonded with the solid phase or the enzyme activity of the composite body which does not bond therewith is determined by measuring the absorbancy of the coloring generated by adding a substrate H2O2 soln. for the enzyme and o-phenylene diamine to the sample, by which a trace amt. of HCG is quantitatively determined with high accuracy.
