ANTI-INTERFERON MONOCLONAL ANTIBODY, AND PURIFICATION OF HUMAN LEUKOCYTE INTERFERON USING SAID ANTIBODY

摘要

PURPOSE:To provide a monoclonal antibody capable of bonding with the molecule of HuIFN-alpha and neutralizing the activity of the considerable portion of the molecule, and effective for the efficient purification of HuIFN-alpha. CONSTITUTION:An anti-HuIFN monoclonal antibody characterized by (i) having specificity to IFN, (ii) produced by hybridoma, (iii) having IgG (H-chain is gamma2b and L-chain is kappa-type) as an isotype, (iv) giving plural interferon-activation molecules within the molecular weight range of 16,400-44,000Dalton when used as an immuno-adsorption using a crosslinked gel as a carrier, reacting with a human leukocyte interferon specimen and eluted with an acidic buffer solution, and (v) giving the activity recovery of 90-100%. The human leukocyte IFN can b epurified up to the purity of 1X108IU/mg-protein in a recovery of 90-100% by the two-stage affinity chromatography using a combination of the monoclonal antibody and a polyclonal antibody originated from different kind of animal.
