The formula involved in the culture medium is as follows: Proteose Peptone No.3 (Difco) (RTM) 38 grams, starch (1252 Merck) (RTM) 25 grams, sodium chloride 3 grams, trimethoprimlactate 15 milligrams, thioglycolate 1.1 grams, horse serum 100 millilitres, 0.06 M sodium phosphate buffer pH 7.8 to 1.000 millilitres. Proteose peptone N3 and sodium chloride are dissolved in 200 ml buffer by a careful warming process. Then, other ingredients are added in the following sequence:-trimethoprimlactate buffer, starch (during continuous stirring), 1.1 g thioglycolate and buffer to 900 ml. The vessel is placed in a water bath at 100 deg.C and agitated continuously for 20 mins. It is then removed, shaken and then allowed to cool. The bouillon is then autoclaved at 121 deg.C for 15 mins, and allowed to cool to 56-60 deg.C. At this stage, inactivate horse serium is added. The bouillon is allowed to cool, and is then placed into test tubes at the rate of 1.5 ml per tube.
