A method for determining the presence of a ligand is disclosed which utilizes a reagent which comprises a specific binding substance for the ligand labeled with an avidin reagent covalently bonded thereto and a biotin labeled enzyme reagent. Various protocols may be used to assay the resulting enzyme activity which is related to the amount of ligand.
