Penicillin G or surfactant addition time and concentration in glutamic acid fermn. process are determined by using the proportional relation between the optical density and the total cell number of fermn. broth. Thus, optical d. of fermn. broth is measured at 562nm and 610nm and the injection time and concentration of penicillin G are determined. The yield deviation based on sugars is 0.2%, that of the traditional method is 1.1%.
