PURPOSE:To provide the titled mutant cell strain obtained by culturing a cell separated from a human hepatocarcinoma cell strain, capable of producing considerable amount of surface antigen against hepatitis virus B, culturable in a serum-free medium, and giving an antigen effective as a preventive vaccine to hepatitis B at a low cost. CONSTITUTION:A mutant cell strain huGK-14 capable of producing considerable amount of surface antigen against hepatitis virus B is separated from a human hepatocarcinoma cell strain, and the mutant strain is cultured. The mutant cell strain has a chromosome mode number of 51 and integrated with about 8 DNA fragment containing a gene coding an abr-type hepatitis virus B surface antigen based on one chromosome haploid. The cell is epithelial adhesive cell having irregular circular form and has optimum proliferation temperature of 36.5+ or -1 deg.C. The productivity of antigen increases rapidly after the cell density reaches saturated state. The proliferation of the cell is carried out preferably in a medium obtained by adding 10% serum to a DM-160 base medium, however, excellent proliferation can be attained even in a serum-free Williams E base medium.
