PURPOSE:An enzyme mixture obtained from the culture medium for Aspergillus oryzae is subjected to a chromatography using a specific affinity adsorbent to produce ribonuclease T1 in hihg yield. CONSTITUTION:An enzyme mixture obtained from the culture medium of Aspergillus oryzae, that is' an aqueous solution of takadiastase mainly containing diastase, is purified by subjecting it to a chromatography using an affinity adsorbent of the formula in which 5'-guanosine monophosphate is the ligand, which links with the 2'-C in the ribose at the position of the 3'-C, to give the ribonuclease T1.
