PREPARATION OF HUMAN IGA SECRETORY CELL

摘要

PURPOSE:To obtain human IgA secretory cell in a high yield, by cultivating human B lymphocyte in a specific human lactoferin-containing medium. CONSTITUTION:A solution obtained by dissolving or dialyzing human lactoferin against a buffer solution (e.g. imidazole buffer solution) containing 2-40mM Mg at pH about 5-9 is reacted with a nuclease (e.g. D NaseI) at 5-65 deg.C for 10-20min. Then human B lymphocyte is inoculated to a medium (e.g. medium RPMI 1640) containing 5-50mug/ml of the human lactoferin, cultivated about 30 deg.C for 3-4 days and the human B lymphocyte is differentiated and introduced to IgA secretory cell.
