METHOD FOR TISSUE CULTURE OF SAFFRON CORM

PURPOSE:To efficiently carry out tissue culture of the subject corm having stable quality without contamination by plant diseases in a large amount, by culturing tissual pieces of a corm in a solid culture medium, forming a cormel, transplanting the resultant cormel to a culture medium in a high cytokinin concentration and thickening the cormel. CONSTITUTION:A parent corm of saffron is successively sterilized in 70% ethyl alcohol and 30% sodium hypochlorite and sufficiently washed with sterile water to aseptically cut out tissual pieces from the corm. The resultant tissual pieces are subsequently cultured on a solid culture medium, such as agar culture medium, to form a cormel, which is then transplanted from the solid culture medium to a solid or liquid culture medium in a higher cytokinin concentration and cultured at 25 deg.C in the dark to thickness the cormel. Thereby, the aimed saffron corm of stable quality without contamination by plant diseases is obtained by the culture process in two stages.
