PURPOSE:To enable high-efficient production of human epithelial growth factor(hEGF) by transforming Escherichia coli with a manifestation vector including hEGF of high translation efficiency into the polypeptide, and culturing the transformant. CONSTITUTION:A hEGF gene of high translation rate into the corresponding peptide is chemically synthesized and the gene is bonded to the manifestation vector which is stable in the cell bodies of Escherichia coli and high in manifestation efficiency. Then, the manifestation vector is used to transform Escherichia coli and the transformed microorganism is cultured to efficiently culture hEGF. The manifestation vector is suitably the one having the DNA sequence given in formula I containing the gene coding 53 amino acids in hEGF following the methionine at the N-terminal of the trpE polypeptide.
