PURIFICATION OF ARGININE RESIDUE-SPECIFIC ENDOPEPTIDASE

PURPOSE:To purify an arginine residue-specific endopeptidase in high yield, by adsorbing an arginine residue-specific endopeptidase to a benzamidine- immobilized carrier to remove foreign matters and eluting the adsorbed enzyme. CONSTITUTION:An arginine residue-specific endopeptidase is made to contact with a benzamidine-immobilized carrier in a 0.01-0.1M phosphoric acid buffer solution of pH7-8 to effect the adsorption of the arginine residue-specific endopeptidase and remove foreign matters. The adsorbed enzyme is eluted with a buffer solution produced by adding about 1M of sodium chloride to the above buffer solution and then eluted with an acid solution of pH2-3, e.g. about 1mM of hydrochloric acid. The purified arginine residue-specific endopeptidase exhibits single band in polyacrylamide electrophoresis.
