In the method, using the synthetic method of solid phosphite the oligonucleotide coding for human insulin A and B was synthesized. From the synthesized oligonucleotides, the genes for A and B chain of insulin was constructed by the shot-gun method. After the E. coli plasmid, pUC19 was opened by the restriction enzyme Sph I and BamH I, and the above constructed gene was inserted by the reaction of DNA ligase. The inserted plasmid was called pSY A and pSY B. The base sequence of pSY A and pSY B was identified by the Sanger's method.
