High yield extn. of high mol.wt. DNA from fish roe - by incubating in salt soln. with protease, addn. of phenol, phase sepn. then pptn. with alcohol
Extn. of DNA, esp. from animal organs such as fish soft roe comprises (1) grinding the roe and suspending in a salt soln.; (2) treating with proteolytic enzymes in presence of ionic detergent; (3) treatment in suspension, with phenol to complete lysis of proteins; (4) physical sepn. of the phenol phase (contg. protein decomposition prods.) to recover a DNA-contg. liq. and (5) pptn. of DNA from this with EtOH. Specifically the NaCl content of the salt soln. is not over 1M and the enzyme used is papain. USE/ADVANTAGE - This method is rapid (only 1/6 of the time of conventional processes) and provides good yields of very pure high mol.wt. DNA on a large scale. Successive treatment with enzyme and phenol in a single reactor simplifies the process and produces a synergistic action. The prods. are useful e.g. in cosmetics for regeneration of the skin.
