Culturing atmospheric nitrogen-fixing bacterial primarily on solid substrates to facilitate detection. - Substrates are prepd. in two stages. The base substrate contains (all wt.%): 1) Na2HPO4.12H2O (1.11-1.56); 2) KH2PO4 (0.13-0.18); 3) Dist.H2O. (92.8-96.19); 4) ionic agar (1.45-2.01); and is sterilised pref. at 121 deg.C. - Another soln. is prepd. separately, contg. 5) MgSO4.7H2O (0.011-0.033); 6) CaCl2.2H2O (0.001-0.002); 7) FeCl3 (0.0001-0.001); 8) Na2MoO4.2H2O (0.001-0.0003) and sterilised similarly to the base substrate and then; 9) D-Mannitol sol. (1.11-3.35) sterilised by filtration. Following this, components 9,6,7,8 and 5 (pref. in this order) are added to the base substrate (pref. cooled to 50 deg.C); under thorough agitation and sterile concns. - The solid minimal nutrient prepd. has a pH of 7.2-7.45 pref. 7.35. This minimal, nitrogen-free substrate may be prepd. without ionic agar
