a process for isolation of a mutant strain of escherichia coli with a periplasmic protease has a defective cell gene mutation.coli having: (a) an inner membrane and an outer membrane, (b) a periplasmic space between the membranes, (c) a protein in a first mobile, able to cross the outer membrane and enter the cell into the surrounding environment, the protein in the pr first state can be detected in the surrounding environment, and still in a second state.when it is inside the cell, and (d) a periplasmic protease having the second state to the first state of the protein in the cell. it then chooses a mutant cell that produces a detectable levels of the protein into the surrounding medium is lower than the level produced by the cells of e. coli, the mutant cell including the periplasmic protease defective.the invention also relates to mutant strains of e. coli with a periplasmic protease defective.
