The invention relates to a lysis reagent for the determination of at least three leukocyte populations including eosinophiles by volumetric differentiation: lysis reagent which comprises a mixture of an aqueous solution of a quaternary ammonium salt, of at least one tertiary ammonium salt, of one leukocyte-protecting agent of the type derived from adamantane and, optionally, of an alkali metal cyanide to convert haemoglobin to a chromogen.
