PURPOSE:To obtain a diagnostic of a human adult T cell leukemia virus (HTLV-1) at large quantities by culturing a hybridoma which is a fused cell obtained by a specific immune. CONSTITUTION:A surface antigen gene of human leukemia virus is cloned to afford plasmid pKHe (A). Then plasmid p7.5-18 (B) derived from vaccine virus (W)WR strain is coupled to the ingredient A to give plasmid p7.5env (C), which is then coupled to a plasmid pHA13 (D) obtained by purifying virus of WWR strain to provide pHA7.5env (E). Then the ingredient E is introduced into a virus-infected cell to provide a recombinant virus WR-7.5env-1 (F). A rat is immunized with the ingredient F and successively additionally immunized with the inactivated ingredient F to give a spleen cell, which is fused with myeloma cell to provide a fused cell HTLV-MAb1 (G). Then the G cell is cultured in mouse ascites to produce the monoclonal antibody capable of specifically reacting with HTLV-1 outer covered protein gp46.
