PURPOSE: To produce a N-acyl-L-proline-acylase by aerobically culturing N-acyl- L-proline with an aqueous culture medium containing Comamonas testosteroni DSM 5416, etc., and having a specific pH value. ;CONSTITUTION: An N-acyl-L-proline-acylase is obtained by preparing an aqueous culture medium by bringing the medium to contain sources of carbon and nitrogen, mineral salts, complex vitamin sources, Comamonas testosteroni DSM 5416 or Alcaligenes denitrificans DSM 5417 and adjusting the pH to 6.0-8.0, adding N-acyl-L-proline to the aqueous culture medium and aerobically culturing at 25-35°C, then, separating cellular materials and successively isolating the enzyme.
