PURPOSE:To extract and immobilize a single stranded DNA automatically, accurately and in good reproducibility by using a system of controlling temperature, time, etc., by a microcomputer, etc. CONSTITUTION:In a process wherein a material inoculated with plaque by phage or colony by bacteria (hereinafter prescribed as target microorganism) or one sheet or plural sheets 2-23 of a target microorganism itself held on a supporting film are successively immersed in a solution to degenerate a transferred material thereof, a second solution to neutralize the solution, a solution to purify DNA and plural kinds of cleaning solutions 2-1 to 2-8 in a treating tank 2-25 comprising one tank, single stranded DNA is separated and immobilized to the supporting film, reaction is attained successively by devices (controller 2-20 and CPU 2-21) to control kind of reaction solutions 2-1 to 2-8 and 2-27 to be sent to the tank 2-25, total flow rate, time interval maintained and continued in the tank 2-25 and temperature in the tank 2-25 according to signals set by a user.
