Procedure for preparation of synthetic oligonucleotides, for gene amplification of specific DNA molecular probes for the male genome of ruminants. It consists in the fact that the oligonucleotides are provided with fragments of a molecular probe which comprises a nucleotide sequence of 49 base pairs of the following formula (A): IMAGE which is already known and which constitutes a DNA segment specific for the male genome of ruminant mammals, especially of the Bos genus, the fragments of which are characterized by comprising at least 10 bases of said sequence A, among which are at least 5 consecutive bases of the latter, as well as their complementary fragments or any fragment which has at least 60% homology with said oligonucleotides. Procedure for gene amplification of specific DNA molecular probes for the male genome of ruminant mammals, which have one of said sequences, by the fixing, by means of hybridization at each of its ends, of a flanking sequence selected from the above- mentioned fragments, and subsequent enzymatic extension and denaturation process.
