PURPOSE:To obtain the subject soft callus capable of simply carrying out enzymic treatment for isolating protoplasts without any differentiation with light by culturing explants containing meristems of Rosmarinus officinalis L. using a culture medium containing auxin for culturing plants in the dark. CONSTITUTION:Laminas are used as explants containing meristems of Rosmarinus officinalis L. which is a perennial arborous plant of a species belonging to the family Labiatae are used, sterilized in 70% ethanol for 3min, then disinfected with a 1% solution of sodium hypochlorite for 20min, washed with distilled water and subsequently cultured in a germ-free state using a culture medium containing auxin which is a phytohormone for culturing plants in the dark to provide a callus of the Rosmarinus officinalis L. Furthermore, the resultant callus is treated with an enzymic solution containing a cellulase, a polygalacturonase, etc., to afford protoplasts of the Rosmarinus officinalis L.
