CONVERSION TO ACTIVATED HUMAN MOTILIN ANALOG AND PURIFICATION OF HUMAN MOTILIN ANALOG

PURPOSE:To obtain the subject substance useful as high-purity pharmaceuticals, etc., on an industrial scale at a low cost with remarkably shortened purification step by treating an inclusion corpuscles produced in a transformed microorganism with acetone, disintegrating insoluble fraction, treating with formic acid and activating with cyanogen bromide. CONSTITUTION:Insoluble inclusion corpuscles produced in the transformant cell of E.coli, etc., as a fused protein consisting of a multimolecular linked product of human motilin analog is solubilized by sterilizing the produced E.coli with acetone, removing the acetone-soluble substances, disintegrating the insoluble fraction and treating the fraction with a solution containing formic acid. The solubilized product is treated with cyanogen bromide and converted to active human motilin analog. The solution of the analog is neutralized with an alkali, the impurities are removed by salting-out and the supernatant is purified by successive hydrophobic chromatography and reverse phase chromatography to obtain the objective activated human motilin analog.
