CULTURE MEDIUM IN TISSUE CULTIVATION OF PLANT AND PRODUCTION THEREOF

PURPOSE:To smoothly grow a plant without causing drying and breaking of a solid culture medium in a vessel, by coating the surface of the solid culture medium for tissue cultivation of the plant with a wax so as to leave only a part required for growing of the plant. CONSTITUTION:The surface of an agar culture medium for carrying out tissue cultivation of a plant is masked at a part required for growing the plant, particularly the central part and a wax is cast on the remaining surface of the solid culture medium to uniformly coat the whole area of the surface at a given width. The uncoated part is preferably 25-50% area in the central part based on the surface area of the above-mentioned culture medium. A cylindrical vessel is preferably used. The wax at 1/3-4 weight ratio of the paraffin/vaseline is initially present in the vessel and the solid culture medium used for the tissue cultivation in a liquid state is cast on the afore-mentioned vessel, sterilized while being kept at 100 deg.C for about 1hr and 120 deg.C for about 15min and allowed to cool. Thereby only the peripheral part is coated with the wax in the form of a doughnut so as to leave only the central part of the culture medium.
