摘要
PURPOSE:To obtain the subject peptide, having a specific amino acid sequence, capable of inducing a viral neutralizing antibody on Aujeszky's disease virus gIII and useful for designing, etc., a peptide vaccine effective in phylaxis of the Aujeszky's disease virus. CONSTITUTION:A virus is purified from Aujeszly's disease virus Indiana S strain according to a conventional method, treated with proteinase K and then with phenol-chloroform to provide a virus DNA. Two primers are bound to both terminals of the resultant gIII gene according to a base sequence of the gIII gene in Aujeszky's disease virus Becker strain and amplified by a polymerase chain reactional(PCR) method. The prepared sample after the amplification is subjected to agarose electrophoresis to cut out a band having a length of the gIII gene. The obtained band is subsequently bound to a vector to afford a recombinant plasmid, which is then used to transform Escherichia coli. The prepared transformant is cultured in a culture medium to provide the objective peptide constructed from an amino acid sequence expressed by the formula.
