PURPOSE: To obtain the subject cDNA, having a specific base sequence, capable of coding a transit peptide of the β-subunit in a rice mitochiondrial ATPase and recovering the sterility of male sterile rice plant, and useful for producing hybrid rice, etc. ;CONSTITUTION: The whole RNA is extracted from a ripened seed of rice plant NIHONBARE (a strain of rice) according to a lithium chloride method and then subjected to oligodT cellulose column chromatography to isolate an mRNA, which is then used as a template to synthesize cDNA. The resultant cDNA is used to prepare a cDNA library and two kinds of primers are subsequently synthesized from a base sequence of the β-subunit in corn mitochondrial ATPase and used to synthesize a partial DNA according to a polymerase chain reaction(PCR) method. The prepared partial DNA is used as a probe to clone the library and select a positive clone. The DNA is then recovered from the positive clone and treated with a restriction enzyme to afford the objective cDNA capable of coding a protein of the β-subunit in the rice mitochondrial ATPase.;COPYRIGHT: (C)1993,JPO&Japio
