PURPOSE: To provide the subject new sequence capable of preparing a manifestation vector having various manifestation powers such as a manifestation vector having strong manifestation power to be used for the production of protein or a vector for the determination of promoter activity. ;CONSTITUTION: A modified upstream activation sequence (UAS) having the structure of yeast glycerol-3-phosphate dehydrogenase 3 (TDH3) gene containing partially depleted natural UAS. The modified UAS can be produced e.g. by producing a plasmid pXX from a TDH3 gene separated from the chromosome of a yeast AH22, digesting the plasmid with Xbal and SphI, subjecting to depleting treatment with ExoIII nuclease, cyclizing the plasmid with T4 ligase, transforming an E.coli with the cyclized plasmid, selecting a cell containing a plasmid having depletion mutation from the colony of the E.coli and preparing a single stranded DNA from the colony.;COPYRIGHT: (C)1993,JPO&Japio
