New human interferon gamma (HIFNG) monoclonal antibodies - produced by immunising mice with spleen cells from mice having high serum antibody titre for HIFNG, followed by immobilised HIFNG
Prodn. of human interferon-gamma monoclonal antibodies (hIFNg-MABS) comprises (i) immunising Balb/C mice with spleen cells of another mouse having a high antibody (AB) titre, (ii) further immunising (a) with HIFNg immobilised on nitrocellulose, (iii) fusing spleen cells of (b) with myeloma cells, (iv) and selecting hybridomas by ELISA having at least a five fold extinction compared to a standard. Pref. the selection is with HIFNg bound to a microtiter plate with polyclonal HuIFNg-specific mammalian AB and detection of bound MABs with anti-mouse ABs. USE/ADVANTAGE - The new ABS may be used in a kit for the determn. of huIFNg and in columns for immuno-purificn. of hIFNg. Using spleen cells from another mouse, less animal material is needed and the experimental time is shortened. Compared to known ABs, the current ABs have a higher affinity.
