PURPOSE:The gene of Bacillus amyloliquifaciens which secretes a large quantity of neutral protease in a short time is utilized to effect efficient secretion of protein. CONSTITUTION:The DNA sequence is prepared, which originates from the neutral protease in Bacillus amyloliquifaciens and contains the promotor region, the ribosome region and the region relating to protein secretion. The preparation is carred out, e.g., by cleaving the chromosome from Bacillus amyloliquifaciens with restriction enzyme, Sau3AI, and connecting it to plasmid PUB110. An example of the resultant DNA sequences is given, where the sequence from No.179 to No.184 shows -35 region, that from No.203 to No.209, -10 region; from 236 to 243, ribosome bonding region; from No.251 to No.913, the region participating the secretion and from No.914 to N0.919, the sequence coding the amino-terminal outside the cell body. Thus, a desired foreign gene is bound to the down stream to enable high-efficiency secretion of the objective substance outside cell bodies.
