PURPOSE: To extract DNA with a high yield and a high purity with a simple operation quickly by allowing an aqueous solution including at least one type selected from a group consisting of protein denaturant and a reducer and then a surface-active agent and chelator to be made of a protein dissolution agent. ;CONSTITUTION: The title DNA extraction reagent is an aqueous solution including a protein denaturant as an essential ingredient. A chaotropic agent is desirable as the protein modification agent. Further, the reagent includes at least one type selected from a group consisting of a reducer, a surface-active agent, and chelator. Thiol reducer which can cut disulfide coupling, one which does not cause settlement in the aqueous solution with a reagent composition, and one which has a capacity for chelating divalent metal ion are desirable as the reducer, the surface-active agent, and chelator, respectively.
