IDENTIFICATION OF REAGENT COMPOSITION AND RETICULOCYTE IN WHOLE BLOOD AND USE THEREOF FOR INDICATING CHARACTERISTICS

PURPOSE: To discriminate a reticulocyte in a blood sample better from other cells using a reagent composition suitable for fluorescent conduction type cell measurement. ;CONSTITUTION: A suspension is formed by mixing an aliquot in a blood sample with a reagent composition containing an organic cation dye compound for dyeing the ribonucleic acid of reticulocyte, a globing agent, and a buffer solution for sustaining pH 6-9. The suspension is subjected, cell by cell, to optical illumination with exciting wavelength in the red or blue region of spectrum and the scattering light from each cell is detected along with fluorescence therefrom. Dyed and undyed cells are then identified based on the scattering light and fluorescence and then the number and volume of reticulocyte or erythrocyte are measured along with the concentrations of hemoglobin and RNA. Finally, the reticulocyte and the erythrocyte in the whole blood sample are discriminated and the characteristics thereof are indicated.;COPYRIGHT: (C)1994,JPO
