CAPILLARY COLUMN FOR SEPARATION AND DETECTION IN ELECTROPHORESIS

PURPOSE: To improve sensitivity and selectivity in the detection of on column ultraviolet detection by applying a coated layer on an internal wall of a capillary column to fill a cavity with an ultraviolet ray hydrophilic polymer network. ;CONSTITUTION: A capillary column contains a capillary 10, a coated layer 16 of a coating material applied on the internal surface 14 of the side wall of the capillary 10 and an ultraviolet ray permeable hydrophilic polymer network material 12 filled into a bore of the capillary. The capillary column containing the ultraviolet ray permeable polymer network enables maintaining of the polymer network therein even under coarse electrophoresis conditions that allows using of a broad range of an electrophoresis electric field containing a relatively high electric field ranging from about 50V/cm to about 100V/cm. Hence, this column is especially suitable for capillary electrophoresis. The capillary column is also especially for the measurement of an SDS protein at a relatively low ultraviolet wavelength of about 214nm (nanometer). The detection at such a wavelength is very sensitive as compared with that at 280nm and more selective.;COPYRIGHT: (C)1993,JPO
