PURPOSE: To provide a novel gene containing a DNA coding uricase originated from Candida.utilis and capable of efficiently producing the uricase useful for the determination of uric acid in blood or urine without adding the uric acid to a medium. ;CONSTITUTION: The novel uricase gene coding an amino acid sequence of the formula is obtained by inoculating Candida.utilis ATCC 9950 (ATCC 74151) on a medium, subjecting to a vibration culture at 30°C for 40 hr, centrifuging the cultured product, suspending the collected cells in a sorbitol solution by a conventional method, recovering the chromosomal DNA, treating the recovered DNA with a restriction enzyme to form a DNA library combined with a phage vector, screening the library by a plaque hybridization method using a probe comprising a base sequence coding the amino acid sequence of a fragment of the uricase, collecting the DNA from the screened positive clone, and subsequently treating the DNA with a restriction enzyme.;COPYRIGHT: (C)1993,JPO&Japio
