PURPOSE: To obtain a new α-L-fucosidase useful for structural analysis, etc., of a sugar chain in a complex carbohydrate. ;CONSTITUTION: This α-L-fucosidase is capable of isolating L-fucose by acting on a natural polymer substrate such as oligosaccharide and α-L-fucosyl bond in a synthetic substrate. The enzyme reacts with α→1→2 or α→1→6 fucosyl bond in a natural substrate and not reacts with α→1→3 or α→1→4 fucosyl bond in the natural substrte, and has optimum pH at 7.0-8.0, stable pH at 6.0-9.0 (at 45°C for 1hr), optimum temperature at 55°C, and when the enzyme is retained at pH7.0 for 10min, the enzyme is stable until 50°C while the enzyme is deactivated at ≥65°C. The enzyme has the molecular weight of about 102,000 (PAGE) or about 51,000 (SDS-PAGE). The enzyme is inhibited by Cu, Hg and para- chloromethacryl benzoic acid, but hardly influenced with L-fucose.
