SUBSTANCE: new strain of bacterium Bacillus stearothermophilus ВКПМ B-5465 is proposed a producer of restriction endonuclease which can recognize and cleave nucleotide sequence 5′-CC(A/T)-GG-3′. Strain is isolated from thermal water sources as a result of restrictase producer screening. The yield of enzyme is 100000 U/g wet biomass, specific activity of the end enzyme is 30000 U/ml. Restrictase Bst 381 is an isoschizomer of restrictase Bst GII and can replace the latter in all genetic engineering studies.
