The derivatisation of amino acids using phenyl isothiocyanates for HPLC analysis can be greatly simplified by the removal of excess phenyl isothiocyanate after addition of a suitable aqueous solution for phase separation and removal of the phenyl isothiocyanate-containing lipophilic phase by filtration through a suitable membrane filter or adsorption of the lipophilic phase on a suitable material. The previously customary complicated vacuum drying of the amino acid derivatives is thus unnecessary; the derivatisation of amino acids with phenyl isothiocyanates can now be readily automated.
