PURPOSE:To obtain the subject improver, containing a human urinary kininogenase extracted and purified from human urine and having a specific molecular weight as an active ingredient and further citric acids, capable of dilating fine cerebral blood vessels and increasing the cerebral blood vessels and useful for preventing and treating, etc., cerebral dysfunction. CONSTITUTION:A cerebral function improver contains a human urinary kininogenase prepared by adding hydrochloric acid to healthy adult male urine, regulating the pH to 5, adding chitosan to the regulated urine, stirring the resultant mixture for 1 hr, adsorbing kininogenase in the urine on the chitosan, filtering the adsorbent, washing the adsorbent with water, eluting the kininogenase with a 0.2M tris(hydroxymethylamino)methane (tris)-hydrochloric acid buffer solution (pH 9.0), filtering the eluate, providing an extract solution, dialyzing the obtained extract solution with a 0.1M sodium hydrogencarbonate buffer solution (pH 9.0) containing 0.5M NaCl, treating the dialyzate with a column in which aprotinin is immobilized and eluting the adsorbed substance with a 0.1M acetic acid buffer solution (pH 3.5) as an active ingredient.
