PURPOSE: To obtain a spacer DNA to be inserted into a manifestation vector, capable of readily establishing a cell strain to highly produce a desired protein without using a specific host cell strain and to prepare the manifestation vector into which the spacer DNA is inserted. ;CONSTITUTION: In a method for transducing a manifestation vector for an eucaryocyte, containing a first DNA sequence coding a desired protein and a second DNA sequence as a marker gene, having an inner ribosome recognition site at the 5' terminal side of the second DNA sequence, into a host eucaryocyte and amplifying the second DNA sequence and the number of copies of the first DNA sequence, a spacer DNA, which is at least not partially translated into a protein or has divergence between the second DNA sequence and the reading frame even in such a case that the base sequence is totally translated, is inserted into a space between an ATG sequence coding an AUG sequence which can be recognized by the ribosome of an eucaryote and an initiating AGE sequence of the second DNA sequence to reduce translation efficiency of the second DNA.;COPYRIGHT: (C)1996,JPO
