MONOCLONAL ANTIBODY SPECIFICALLY REACTING WITH INTRACELLULAR DOMAIN OF C-ERBB-2 GENE PRODUCT AND HYBRIDOMA SECRETING THE SAME ANTIBODY

PURPOSE: To obtain a new monoclonal antibody specifically reacting with a intracellular domain of c-erbB-2 gene product, not reacting with a receptor of human epithelium growth factor, and useful for diagnosis and curing, etc., of a cancer in an immunological method. ;CONSTITUTION: A plasmid containing cDNA of a human c-erbB-2 gene as one kind of cancer gene is treated with a restriction endonuclease, the treated c- erbB-2 gene is cut out and is integrated into an expression vector and manifested in a host cell to obtain a c-erbB-2 gene product. Next, the product is injected into a mouse cavum abdominis together with an adjuvant and immunized, then a lien cell is taken out after the last immunization and fused with a mouse myeloma cell, and selectively cultured in a HAT medium to make a hybridoma, an antibody-forming stock is selected and cloned by a limiting dilution method, then cultured and specifically reacted with an intracellular domain of a c-erbB-2 gene product to obtain the objective monoclonal antibody not reacting with a receptor of human epithelium growth factor.;COPYRIGHT: (C)1996,JPO
