PURPOSE: To obtain a promoter capable of giving an enzyme effectively, useful for decomposing and removing raffinose in a beet sugar production, by extracting from Saccharomyces oleaginosus as a promoter for α-galactosidase gene. ;CONSTITUTION: A chromosomal DNA is obtained by inoculating Saccharomyces oleaginosus IFO 1998 strain on a medium, after culturing it with shaking at 30°C for about 36hrs., collecting microbes by an centrifuge of the culture medium, suspending obtained wet microbes in a buffer solution, bacteriolysing by adding an enzyme for cell wall lysis, further treating by adding a ribonuclease and a pronase, and by precipitating with ethanol after treating with phenol and chloroform. Next, this promoter having a base sequence shown in the formula, three TATA boxes and one UAS region, is obtained by treating the above chromosomal DNA with a restricting enzyme, fractionating by an agarose electrophoresis, introducing to a vector for transformation of a host, culturing the host to select a strain capable of expressing an α-galactosidase, and recovering a DNA to treat with a restriction enzyme.;COPYRIGHT: (C)1995,JPO
