EXPRESSION OF PERTUSSIS HOLOTOXIN IN BORDETELLA PERTUSSIS

PURPOSE: To obtain the subject plasmid construct containing a pertussis holotoxin gene located at the 3'-position of the ptx operon encoding factor of a Bordetella pertussis genome and capable of producing the pertussis holotoxin for the production or the like of a pertussis vaccine in a heterologous host on a large scale. ;CONSTITUTION: This new plasmid construct contains a base sequence represented by the formula and further a Bordetella pertussis nucleic acid sequence of about 8 kb located at the 3'-position of a ptx operon encoding factor of the Bordetella pertussis genome causing pertussis or acute infectious diseases in the respiratory tract and capable of encoding a gene product required for the expression or secretion of Bordetella pertussis holotoxin. The plasmid construct is transferred into a heterologous host to transform the host. The resultant transformant is then kept under conditions suitable for expression and secretion of the ptx to thereby produce the pertussis holotoxin required for the production of a pertussis vaccine on a large scale.;COPYRIGHT: (C)1995,JPO
