The present invention relates to generation of specific antigens associated with P. multocida under controlled culture conditions and the use of such conditions in monitoring the generation of specific antigens. The invention relates to antigens preferably derived from culture supernatant, and most preferably the invention relates to identifying and purifying antigens from culture supernatant of a culture of P. multocida. The present invenltion also relates to these antigens and the use of such antigens in the detection and purification of a number of protective and diagnostic antigens and antibodies from parasites and particularly bacteria. In particular, the present invention relates to the identification and purification of antigens in the bacterium family Pasteurellae, preferably Pasteurella multocida, and most preferably Pasteurella multocida type A:3. The invention also relates to the use of the antigens in vaccines and in assays for diagnostic purposes. The vaccines of the invention have antigens produced in accordance with the invention, preferably the vaccines have antigens derived from a culture supernatant of a culture of bacterium growth under controlled conditions, and more preferably, growth under iron-restricted culture conditions.
