A method, device and kit for determining total protein concentration in a protein solution, which comprises applying the protein solution to a protein-support, wherein the protein-support is capable of retaining the total protein in the form of a protein spot having a size that is proportional to the protein concentration when the protein contacts the protein-support in the presence of a detergent and at least one of a salt or an acidic agent; measuring the size of the protein spot produced on the protein support with a gauge, such as is depicted in the figure for example; and comparing the size of the protein spot with a standard, also depicted in the figure (see the calibrated markings beneath the circles).
