PROBLEM TO BE SOLVED: To obtain a novel nucleic acid which includes a specific DNA sequence or a part thereof, is used in the amplification of the β-subunit gene of Bacillus tuberculosis RNA polymerase and is useful for detection of chemical tolerance, an important index for selecting the therapeutic agent for tuberculosis. ;SOLUTION: This novel oligonucleotide includes nucleic acid sequence of at least 15 sequential residues selected from the nucleic acid sequences given in formula I or formula II or their complementary DNA sequences and is used in amplification of the β-subunit gene of Mycobacterium tuberculosis. The gene fragment of RNA polymerase β-subunit containing the area causing the mutation obtaining resistance to rifampicin in high frequency is specifically, simply, rapidly and steadily amplified and is useful in the examination on the drug resistance. This oligonucleotide may be prepared on the basis of gene information on Escherichia coli or the like relating to chemical resistance.;COPYRIGHT: (C)1997,JPO
