PROBLEM TO BE SOLVED: To obtain a kaurene synthetase, having a specific amino acid sequence, having catalyzing actions on the conversion of copalyl pyrophosphate into an ent-kaurene which is an intermediate for biosynthesis of gibberellin and useful for regulation, etc., of plant growth. ;SOLUTION: This kaurene synthetase comprises a new polypeptide specified by an amino acid sequence (amino acid Nos. 1-789) represented by the formula, has catalyzing actions on the conversion of copalyl phosphate into an ent- kaurene which is an intermediate for biosynthesis of gibberellin and is useful for regulation, etc., of plant growth. The kaurene synthetase is obtained by cultivating a Cucurbita maxima L., harvesting an immature fruit at the time of growing cotyledons to about half the final length thereof, extracting an endosperm in a seed thereof with a phosphoric acid buffer, etc., then carrying out the gel filtration of a crude enzyme extract solution, successively performing anion exchange chromatography, hydroxyapatite column chromatography, etc., of an active fraction and then purifying the active fraction.;COPYRIGHT: (C)1997,JPO
