FIELD: genetic engineering, medicinal genetics. SUBSTANCE: new recombinant DNA plasmid exhibits human genome DNA fragment (680 base pairs) containing two internal unique restriction site PstI and confined with Eco RI-sites from the different ends of genome fragment. Also, this new DNA-probe has "minor" hybridization sites on human chromosome under conditions in situ that involve chromosomes 2, 4, 14, 15, 18, 20, 22. Under special conditions this DNA-probe is hybridized only with centromeric regions of human chromosomes 13 and 21. So, invention can be used for effective identification of chromosomes 13 and 21 in interphase and metaphase cells for prenatal diagnosis and in clinic cytogenetics in patients with chromosome pathology (Down's and Patau's syndrome). EFFECT: improved method of diagnosis and identification.
